Labeobarbus gananensis was described by the Italian ichthyologist Decio Vinciguerra in 1895 from the Genale River of Ethiopia — the epithet directly references the Genale (or Ganale) drainage. The valid combination follows the large African barb genus Labeobarbus, which unifies the yellowfishes, large river barbs, and closely related groups across sub-Saharan Africa. Prior to modern molecular and morphological revisionary work, many African cyprinids now assigned to Labeobarbus were shuffled between Barbus, Varicorhinus, and regional synonyms; the current Catalog of Fishes placement in Labeobarbus is the accepted valid name.
Phylogenetic studies of Ethiopian Labeobarbus using mitochondrial DNA have confirmed that the Genale River complex constitutes a monophyletic lineage — all six mouth-form variants within the river share a single common ancestor, pointing to in-situ diversification rather than multiple colonisation events.
