Acestrocephalus acutus was described by the Brazilian ichthyologist Naércio Aquino Menezes in 2006 in a paper published in Neotropical Ichthyology, alongside four other new species of the genus — A. maculosus, A. stigmatus, A. nigrifasciatus, and A. pallidus — and redescriptions of A. sardina and A. boehlkei. The type locality is the rio Tocantins drainage of Brazil. The Catalog of Fishes records the valid name as Acestrocephalus acutus Menezes, 2006.
Acestrocephalus belongs to the family Characidae and is placed within a predatory guild of slender, large-toothed characins historically treated under the subfamily Acestrorhynchinae. The genus was established by Carl H. Eigenmann and encompasses a set of stream-dwelling piscivores distributed across Amazonian and Plata drainages of South America. Menezes's 2006 revision provided the most thorough modern treatment of the group, clarifying species boundaries and diagnosing the five new species on the basis of meristic counts, colour pattern, and head morphology.
