Peckoltia multispinis was originally described by Holly in 1929, with the account published in Anzeiger der Akademie der Wissenschaften in Wien, Mathematisch-Naturwissenschaftliche Klasse (v. 66, pp. 119 [3]). The type material originated from the fish market (Mercado) in Belém, Brazil — a market specimen without a precise collection locality, which introduces an inherent ambiguity about the exact provenance and has contributed to ongoing taxonomic uncertainty.
The species has passed through multiple generic assignments over its nomenclatural history; earlier combinations include Ancistrus multispinis and Lasiancistrus multispinis before the current placement in Peckoltia.
Critically, the taxonomic status of P. multispinis is not settled. Armbruster's 2008 monograph on the genus Peckoltia (Zootaxa 1822: 34) raised the possibility that this species is a junior synonym of Peckoltia vittata (Steindachner, 1881). FishBase explicitly flags this uncertainty, stating that the species is either valid as Peckoltia multispinis (Holly, 1929) or a synonym of P. vittata, pending formal resolution. The IUCN Red List currently treats it as a valid species for assessment purposes. The lower Amazon and lower Tocantins are large, complex river systems with diverse habitat types including main-channel sandy shores, blackwater floodplain lakes, and clear rocky sections in the Tocantins gorge area.
