Enteromius lukusiensis was described in 1937 by David and Poll from specimens collected in the Lukusi River, a tributary within the Congo Basin of the Democratic Republic of the Congo. The original genus was Barbus, the catch-all container for African and Asian barbs used through much of the twentieth century. The Catalog of Fishes (Eschmeyer, CAS), the authority for valid names, now places the species as Enteromius lukusiensis (David & Poll, 1937), the parenthetical authority indicating the move out of Barbus. Some older literature and databases may still list the species under the synonymous combination Barbus lukusiensis.
Enteromius is a large African genus erected to accommodate the many small, often single-river barb species formerly lumped in Barbus sensu lato. Revisions of the Congo and central African barb fauna have progressively assigned species to Enteromius on molecular and morphological grounds. The species is listed on FishBase under ID 11395. No subspecies or infraspecific taxa are recognised.
