Labeobarbus iturii was first described by Max Holly in 1929 under the original combination Barbus iturii, based on a type specimen from the Ituri River in what was then the Belgian Congo. The parenthetical authority in the current name — (Holly, 1929) — indicates that the species has been transferred out of its original genus.
The genus Labeobarbus encompasses the large African barbs and yellowfishes, a diverse lineage within the family Cyprinidae, placed in the subfamily Torinae alongside the Asian mahseers. Broader revisions of African Cyprinidae have progressively moved many species formerly lodged in the catch-all genus Barbus into more natural groupings; Labeobarbus iturii sits within that revised framework.
